以烟草叶片为原料,用正交试验法优化了烟草中辅酶Q10的匀浆提取工艺,考察了匀浆时间、乙醇体积分数、液料比和提取次数4个因素对辅酶Q10提取率的影响,确立了烟草中辅酶Q10的优化匀浆提取工艺条件为:提取溶剂为85%乙醇,匀浆时间6min,液料比9∶1(mL/g),提取次数2次.将该法与索氏提取进行了对比.结果表明,匀浆提取烟草中辅酶Q10的提取率为12.82μg·g-1,与索氏提取的提取率相当,但匀浆提取所用提取时间短,提取溶剂用量少,因此匀浆法具有明显的优势.

研究盐胁迫浓度与大麦幼苗生长分裂的关系,实验结果表明:大麦幼苗生长在一定浓度的NaCl溶液中时,幼苗生长抑制、叶绿素含量降低,有丝分裂指数下降,根尖分生区细胞中具有微核和染色体畸变的细胞明显增多.统计分析结果显示:幼苗的分裂指数、遗传损伤与盐浓度间有很好的线性关系,有丝分裂指数与处理浓度间呈负相关(p<0.01),微核率、染色体畸变率两项指标与处理浓度间呈正相关(p<0.01).研究结果表明:大麦幼根有丝分裂指数、微核率及染色体畸变率可以作为监测环境NaCl的定量指标.

系统地研究了中国木犀科苦丁茶ISSR反应体系中的主要影响因子,建立了一套稳定的ISSR-PCR反应参数.筛选出了10个有效引物,并以中国木犀科苦丁茶8个物种共21份种质材料为供试材料对优化后的反应条件的重复性、多态性进行了检测.优化后的反应体系为:10×buffer 2.5 μL,2.0~3.0 mmol·L~(-1) MgCl_2,150~300μmol·L~(-1)dNTPs,Taq酶1.0~1.5 U,引物0.4~0.5 μmol·L~(-1),DNA模板5~320 ng.PCR扩增程序为:94℃预变性4 min,然后按94℃变性40 s,50~5...

莱茵衣藻是研究绿藻产氢的模式物种.为了研究添加外源葡萄糖对莱茵衣藻生长和产氢的影响,本实验通过向莱茵衣藻培养基中加入不同浓度的葡萄糖,检测在光照和黑暗条件下对莱茵衣藻细胞数、叶绿素含量、产氢量和耗氧量的影响,结果表明葡萄糖的添加量为4 g·L~(-1)时衣藻的细胞数和叶绿素含量最高,葡萄糖的添加量为6g·L~(1-)时衣藻产氢量比在正常培养基中的提高6.7倍,达156μL·mg~(-1)chl;但在缺硫培养基中添加外源葡萄糖却没有最终使产氢量增加.结果表明向培养基中加入葡萄糖确实提高了产氢量,为利用衣藻进行含糖废水的处理和产氢的研究奠定了实验基础.
