We have been investigating the role of Fused Toes Homolog (FTS) and its central role in cervical carcinogenesis. Radiation/cisplatin-induced activation of Epidermal growth factor receptor (EGFR) and Notch signaling confers chemo and radio resistance. we identified that FTS is involved in conferring radio- resistance. we identified the regulation of EGFR nuclear Phosphorylation and Notch mediated cancer stem cell activation by FTS upon radiation (x-ray and Carbon ion). Further, silencing of FTS combined X-ray/C-ion/cisplatin combination targets the EGFR, Notch signaling in cervical cancer cells. Also, FTS silencing reduced the spheroid formation ability and cancer stem cells markers (Oct4A, Sox2, Nanog). More importantly, we identified that silencing of FTS reduced the expression of HPV16 E6 and E7 protein levels via ubiquitin mediated proteasomal degradation, and increased the tumor suppressor protein p53 and Rb in HPV 16 positive CaSki and SiHa cells.
